Molecular clues to link between childhood maltreatment and later suicide

ScienceDaily (July 3, 2012) ? Exposure to childhood maltreatment increases the risk for most psychiatric disorders as well as many negative consequences of these conditions. This new study, by Dr. Gustavo Turecki and colleagues at McGill University, Canada, provides important insight into one of the most extreme outcomes, suicide.

"In this study, we expanded our previous work on the epigenetic regulation of the glucocorticoid receptor gene by investigating the impact of severe early-life adversity on DNA methylation," explained Dr. Turecki. The glucocorticoid receptor is important because it is a brain target for the stress hormone cortisol.

The researchers studied brain tissue from people who had committed suicide, some of whom had a history of childhood maltreatment, and compared that tissue to people who had died from other causes. They found that particular variants of the glucocorticoid receptor were less likely to be present in the limbic system, or emotion circuit, of the brain in people who had committed suicide and were maltreated as children compared to the other two groups..

This study also advances the understanding of how the altered pattern of glucocorticoid receptor regulation developed in the maltreated suicide completers. The authors found that the pattern of methylation of the gene coding for the glucocorticoid receptors was altered in those who completed suicide and who also had a history of abuse. These DNA methylation differences were associated with distinct gene expression patterns.

Since methylation is one way that genes are switched on or off for long periods of time, it appears that childhood adversity can produce long-lasting changes in the regulation of a key stress response system that may be associated with increased risk for suicide.

"Preventing suicide is a critical challenge for psychiatry. This study provides important new information about brain changes that may increase the risk of suicide," said Dr. John Krystal, Editor of Biological Psychiatry. "It is striking that early life maltreatment can produce these long-lasting changes in the control of specific genes in the brain. It is also troubling that the consequences of this process can be so dire. Thus, it is important that we continue to study these epigenetic processes that seem to underlie aspects of the lasting consequences of childhood adversity."

While the two biggest stars on the NHL free-agent market were still pondering their futures, a future Hall of Famer found a new home in Texas.

The Dallas Stars signed 40-year Jaromir Jagr to a one-year deal worth $4.55 million on Tuesday.

"He's still a world-class player," Stars general manager Joe Nieuwendyk said. "For us to bring a player like that to our franchise is a big deal."

Ryan Suter and Zach Parise, however, were not yet ready to pick a team to play for next season and beyond.

"When a young man is asked to commit for 13 years, why is it considered extraordinary to take a few days to make a decision?" Neil Sheehy, Suter's agent, wrote in an email. "A deal will happen when it happens."

Parise said Tuesday he still didn't have a timetable for his decision soon after landing in his home state at the Minneapolis-St. Paul International Airport.

"I'm back here to talk with my family and make sure that we've done all the work that we need to do make sure we make the right decision," Parise told the Star Tribune of Minneapolis.

Parise hasn't met with any teams, one of his agents, Wade Arnott, told The Associated Press. His other agent, Don Meehan, said there was no chance the coveted client would make a decision Tuesday night.

"We will resume tomorrow," Arnott wrote in a text message Tuesday night.

The New Jersey Devils want their top forward available to re-sign while Pittsburgh, Philadelphia, Detroit and Minnesota are among the teams that are trying to lure him away.

Parise said he has been looking at where he wants to live, where he'll be happy and what team he wants to play for since hitting the market Sunday. And, he's looking forward to being done with the process.

"It's been a stressful couple days," Parise told the Star Tribune.

The two free agents are both 27 and each has been with only one team during their seven-season careers. Suter has played for the Nashville Predators, who are hoping for a quick answer from him so that they can move onto Plan B if necessary. The Predators aren't alone.

Second-tier free agents such as defenseman Matt Carle and forward Alexander Semin seem to be waiting for Suter and Praise to sign so that they can offer their talents to teams that don't get a top target.

Some players, however, have jumped at chances to re-sign or sign in the opening three days of free agency.

Colorado kept defenseman Erik Johnson thanks to a $15 million, four-year deal and New Jersey retained defenseman Bryce Salvador with a $9.5 million, three-year contract.

Jagr, who played last season with Philadelphia, has led the NHL in scoring five times and won an MVP. He had 19 goals and 54 points ? both career lows ? for the Flyers.

In NHL history, he ranks first with 16 overtime goals, second with 114 game-winning goals, eighth with 1,653 career points, 11th with 665 goals and 12th with 988 assists.

Jiri Hudler signed a $16 million, four-year contract with Calgary ? getting much more than Detroit offered to keep him ? and former Flames forward Olli Jokinen left to play in Winnipeg on a $9 million, two-year contract on Monday.

There were a flurry of signings Sunday ? Day 1 of NHL free agency ? including forward P.A. Parenteau leaving the New York Islanders for a $16 million, four-year contract with the Avalanche and 40-year-old forward Ray Whitney getting a $9 million, two-year deal from Dallas.

In Florida fight, Obama and Romney scrap along I-4

ORLANDO, Fla. (AP) ? In the presidential battleground with the biggest prize, Democrat Barack Obama is focused on ratcheting up voter turnout in Florida's university towns, its Hispanic enclaves around Orlando and its Jewish communities in the south. Republican challenger Mitt Romney is working to squeeze as many votes as possible out of north Florida's conservative military bastions, the senior-heavy Gulf Coast and Miami's Cuban community.

But their strategies to energize core supporters overlap in the central Florida swing-voting region that's key to winning the state and its 29 electoral votes. Voters along Interstate 4, which stretches from Tampa Bay to Daytona Beach, will determine the outcome if the race remains close into the fall, as expected. About 45 percent of the state's voters live in that 17-county area.

"Neither party has enough base alone, which is why those persuadable places, particularly along the I-4 corridor, are so important," said Steve Schale, a Democrat who ran Obama's Florida campaign four years ago.

It seems that's usually the case, judging by Florida's track record of hard-fought races and narrow presidential outcomes since the 2000 race landed at the Supreme Court, which then handed the White House to Republican George W. Bush.

Bush won the state again four years later, 52 percent to 47 percent, over Democrat John Kerry. But in 2008, the state sided with Democrats when Obama defeated Republican John McCain, 51 percent to 48 percent.

This year, the stakes are hard to overstate: Obama's re-election is nearly assured should he repeat his 2008 victory in Florida, based on how the states lean now. His standing in Florida is far more precarious than it is in other contested states ? so if he wins Florida, it's likely that he's won in many other states as he looks to cobble together the 270 Electoral College votes it takes to win. Romney's state-by-state routes to reaching the magic number are more limited than the president's, and a Florida victory would make it far more probable that he could win the presidency.

The electorate in Florida is virtually unchanged from 2008 because the ailing economy stifled the population growth of the previous decade. And in this campaign, the economy dominates.

The recession took a deep toll on the state's recreation industry, especially around Orlando. A decline in foreign trade hurt the Port of Tampa, Florida's largest shipping port. The housing crisis fueled widespread home foreclosures and severely hampered the construction industry on which much of the region's immigrant-heavy workforce relies.

Florida's unemployment rate was 8.6 percent in May, slightly higher than the national average and all other presidential battleground states except Nevada.

A little more than four months before the Nov. 6 election, Obama narrowly leads Romney in statewide polls.

The president and his Democratic allies spent roughly $17 million in television advertising in Florida from April, when Romney effectively became the GOP presidential nominee, through last week. Romney's campaign hasn't been on the air in the state since then, but his allies have doled out $12 million during that time. The ads are heavily concentrated on the Tampa and Orlando media markets, which are cheaper than Miami's and reach far more swing voters.

Of the $2.8 million spent on TV ads in the state last week, $1.8 million was in this region.

Obama is on defense in the I-4 corridor, which he won by a very slim majority in 2008 after Bush won it in 2000 and 2004. Republicans hope that holding their national convention in Tampa in August will give them an edge.

In a close race where anything could be determinative, organization could count hugely and, on this point for now at least, Obama has an advantage. He never dismantled his 2008 campaign infrastructure in the state and has 36 campaign offices. Romney has quickly opened 23, run jointly with the Republican National Committee.

"In a race like this, that is so close and so hard-fought, door-to-door, mail and telephones could make the difference," said Republican Sally Bradshaw, a top Florida aide to Romney during his failed 2008 presidential bid.

Of paramount importance for both candidates right now: energizing and mobilizing core supporters.

For Obama, that means working to tap the burgeoning Puerto Rican community ? an outlier in Florida's sluggish population growth ? around Orlando. Puerto Ricans tend to support Democrats.

Romney's campaign, in turn, sees a chance to shave Obama's advantage among Jewish voters, for whom support for Israel is a top issue. Aides say Romney has tapped into some Jews' irritation over the president's seemingly sympathetic statements toward the Palestinians. Aides said Monday that Romney also planned to visit Israel this summer.

But both candidates stressed central Florida early on.

Obama was in Tampa in April, announcing a measure to promote trade with Latin America. Romney was in neighboring St. Petersburg in May, promoting plans to cut federal spending. Both stopped in Orlando last month to visit businesses and appeal for support from Latino voters.

"The reality is it's the most up-for-grabs part of the most up-for-grabs state," B.J. Neidhardt, manager of Orlando Democrat Val Demings' congressional campaign, said of Florida's midsection.
